1、用途
PAXgene 血液 RNA 系统由采血管（PAXgene 血液 RNA 管）和核酸纯化套件（PAXgene 血液 RNA 套件）组成。它适用于在分子诊断测试使用的逆转录聚合酶链反应中采集、存储和输送血液，在封闭管中稳定细胞内RNA（核糖核酸），随后从全血中分离和纯化细胞内 RNA。

2. 必须使用 BD Vacutainer 采血针固定器，以确保功能正常。参见“订购信息”。

4. 阳性供样者标本识别标签。

B. 标本采集程序

1. 确保 PAXgene 血液 RNA 管使用前在 18-25˚C 温度下，且正确贴有标本识别标签。
2. 如果 PAXgene 血液 RNA 管是待抽吸的唯一试管，应当先抽血至“丢弃管”，再抽血至 PAXgene 血
液 RNA 管，这样便可以灌注放血过程中使用的采血装置的内部空间。否则，PAXgene 血液 RNA 管
应是放血程序中最后抽吸的试管。

3. 采用您所在机构推荐的标准静脉穿刺技术程序，使用采血装置和试管固定器，采血至 PAXgene 血液
RNA 管。应采用以下技术以防止可能出现的血液回流：
a. 将供样者的手臂向下放置。
b. 采血过程中保持试管垂直，处于供样者手臂水平以下。
c. 一旦血液开始流入试管，应松开压血带。
d. 确保试管的附加剂在静脉穿刺期间不会接触到塞子或采血针的末端。
4. 请等待至少 10 秒钟，以便进行完整的抽血。确保血液已停止流入试管再从固定器中取下试管。
PAXgene 血液 RNA 管及其真空经设计可以抽 2.5 ml 血液至试管中。

V. 限制性

VI. 警告和防护措施

5. 丢弃批准处理的生物危害物容器中的所有采血管。

I . Intended Use
The PAXgene Blood RNA System consists of a blood collection tube (PAXgene Blood RNA Tube) and nucleic acid purification kit (PAXgene Blood RNA Kit). It is intended for the collection, storage, and transport of blood and stabilization of intracellular RNA in a closed tube and subsequent isolation and purification of intracellular RNA from whole blood for RT-PCR used in molecular diagnostic testing.
Performance characteristics for the PAXgene Blood RNA System have only been established with FOS and IL1B gene transcripts. The user is responsible for establishing appropriate PAXgene Blood RNA System performance characteristics for other target transcripts .
Summary and Explanation
The collection of whole blood is the first step in many molecular assays used to study intracellular RNA. A major challenge in this type of testing is the instability of intracellular RNA, which rapidly degrades within hours after blood collection. Furthermore, certain species of RNA, through the process of gene induction, increase in vitro after blood collection. Both in vitro RNA degradation and gene induction can lead to an under or overestimation of in vivo relative gene transcript number. The PAXgene Blood RNA Tube contains an additive that stabilizes the in vivo gene transcription profile by reducing in vitro RNA degradation and minimizing gene induction. When used in conjunction with the PAXgene Blood RNA Kit, the PAXgene Blood RNA Tube provides for accurate detection and quantitation of gene transcripts.

Specimen Collection and Preparation for Analysis
A. Required Blood Collection Accessories (Not included with the PAXgene Blood RNA Tube)
Blood collection set such as the BD Vacutainer® Push Button Blood Collection Set or the BD Vacutainer Safety-Lok™ Blood Collection Set. See Ordering Information.

A “Discard Tube” if the PAXgene Blood RNA Tube is the only tube being drawn. See Ordering Information.

Alcohol swab for cleansing site.
Dry sterile gauze.
Tourniquet.
Needle disposal container for used needle or needle/holder combination.
B . Procedure for Specimen Collection

Draw Order:
Tube A - Discard Tube
10 Tube B - PAXgene Blood RNA Tube

Ensure that the PAXgene Blood RNA Tube is at 18−25˚C prior to use and properly labeled with specimen identification.
If the PAXgene Blood RNA Tube is the only tube to be drawn, blood should be drawn into a “Discard Tube” prior to drawing blood into the PAXgene Blood RNA Tube so the interior volume of the blood collection set used during phlebotomy can be primed. Otherwise, the PAXgene Blood RNA Tube should be the last tube drawn in the phlebotomy procedure.

Using a blood collection set and a tube holder, collect blood into the PAXgene Blood RNA Tube using your institution’s recommended procedure for standard venipuncture technique. The following techniques shall be used to prevent possible backflow:
Place donor’s arm in a downward position.
Hold tube in a vertical position, below the donor’s arm during blood collection.
Release tourniquet as soon as blood starts to flow into tube.
Make sure tube additives do not touch stopper or end of the needle during venipuncture.
Allow at least 10 seconds for a complete blood draw to take place. Ensure that the blood has stopped flowing into the tube before removing the tube from the holder. The PAXgene Blood RNA Tube with its vacuum is designed to draw 2.5 ml of blood into the tube.
Immediately after blood collection, gently invert the PAXgene Blood RNA Tube 8–10 times.
Store the PAXgene Blood RNA Tube upright at room temperature (18−25˚C) for a minimum of 2 hours and a maximum of 72 hours before processing or transferring to refrigerator (2−8˚C) or freezer (−20˚C). See Procedure for Freezing and Thawing Specimens in the PAXgene Blood RNA Tubes for details if storage temperature of
−70˚C / −80˚C is desired.
Procedure for Freezing and Thawing Specimens Collected in the PAXgene Blood RNA Tubes
Stand the PAXgene Blood RNA Tube upright in a wire rack. Do not freeze tubes upright in a Styrofoam™ tray as this may cause the tubes to crack.
The PAXgene Blood RNA Tubes can be stored at −20˚C and below. If tubes are to be kept at temperatures below
−20˚C, freeze them first at −20˚C for 24 hours, then transfer them to −70˚C or −80˚C.
Thaw the PAXgene Blood RNA Tubes in a wire rack at ambient temperature (18–25˚C) for approximately two hours. Do not thaw the PAXgene Blood RNA Tubes at temperatures above 25˚C.
Carefully invert the thawed PAXgene Blood RNA Tubes 10 times.
Note: If the two-hour ambient temperature incubation was not carried out prior to freezing, incubate tubes at least another two hours after reaching room temperature before processing.
Note: The frozen PAXgene Blood RNA Tubes are subject to breakage upon impact. To reduce the risk of breakage during shipment, frozen tubes should be treated in the same manner as glass tubes. Users must validate their own freezing and shipping protocol for the PAXgene Blood RNA Tubes.
Procedure for Specimen Preparation for Analysis
Blood samples must be processed according to the instructions provided with the PAXgene Blood RNA Kit.

V . Limitations
Under-filling of the PAXgene Blood RNA Tubes will result in an incorrect blood-to-additive ratio and may lead to incorrect analytic results or poor product performance.
The PAXgene Blood RNA System is not suitable for the collection and purification of viral RNA.
Blood samples obtained with the PAXgene Blood RNA Tube must be prepared only with the PAXgene Blood RNA Kit. For catalog numbers for the PAXgene Blood RNA Kit, see Ordering Information.
The quantity of blood drawn should be 2.5 ml per PAXgene Blood RNA Tube, but this volume might vary with altitude, ambient temperature, barometric pressure, tube age, venous pressure, and filling technique.
The PAXgene Blood RNA Kit is intended for purification of intracellular RNA from human whole blood with leukocytes counts between 4.8–11.0 × 106 leukocytes/ml.
Endotoxin not controlled. Blood and blood components collected and processed in the tube are not intended for infusion or introduction into the human body.

VI . Warnings and Precautions
Contents of this tube are irritating to skin.
After inhalation, supply fresh air: consult doctor in case of complaints.
After skin contact, immediately wash with water and soap, and rinse thoroughly.
After eye contact, rinse opened eye for fifteen minutes under running water, then consult a doctor.
After swallowing, immediately call a doctor.
A blood collection set must be used with the PAXgene Blood RNA Tube. See Ordering Information.
Practice Universal Precautions. Use gloves, gowns, eye protection, other personal protective equipment, and engineering controls to protect from blood splatter, blood leakage, and potential exposure to bloodborne pathogens.
Handle all biologic samples and blood collection devices according to the policies and procedures of your facility. Obtain appropriate medical attention in the event of any exposure to biologic samples (for example, through a needle stick injury), since they may transmit viral hepatitis, HIV (AIDS), or other infectious diseases. Utilize any built-in needle protector, if the blood collection set provides one. PreAnalytiX does not recommend reshielding used needles. However, the policies and procedures of your facility may differ and must always be followed.
Discard all blood collection tubes in biohazard containers approved for their disposal.
Do not re-use the PAXgene Blood RNA Tubes.
Do not use the PAXgene Blood RNA Tubes after the expiration date printed on the tube label.
Since the PAXgene Blood RNA Tube contains a chemical additive, a blood collection set must be used for blood collection to prevent possible backflow from the tube.

Excessive centrifugation speed (over 10,000 RCF) may cause PAXgene Blood RNA Tube breakage, exposure to blood and possible injury.
Do not transfer a sample from a syringe to the PAXgene Blood RNA Tube.
VII . Storage
Store the unused PAXgene Blood RNA Tubes at 18−25˚C. Limited excursion temperatures up to 40˚C are permitted.
Instructions for Removal of BD Hemogard™ Closure

Grasp the PAXgene Blood RNA Tube with one hand, placing the thumb under the BD Hemogard Closure. (For added stability, place arm on solid surface). With the other hand, twist the BD Hemogard Closure while simultaneously pushing up with the thumb of the other hand ONLY UNTIL THE TUBE STOPPER IS LOOSENED.
Move thumb away before lifting closure. DO NOT use thumb to push closure off tube. Caution: If the tube contains blood, an exposure hazard exists. To help prevent injury during closure removal, it is important that the thumb used to push upward on the closure is removed from contact with the tube as soon as the BD Hemogard Closure is loosened.
Lift closure off tube. In the unlikely event of the plastic shield separating from the rubber stopper, DO NOT REASSEMBLE CLOSURE. Carefully remove rubber stopper from tube.
