【背景介绍】免疫球蛋白M(IgM)是由B细胞产生的，迄今为止，是人体循环系统中最大的抗体。IgM是机体受抗原初次刺激后最先产生的抗体。脾脏中的浆母细胞产生抗体，同时脾脏也是特异性IgM产生的主要场所。IgM主要存在于血清中，因为J链的存在，它也是重要的分泌型免疫球蛋白。

 

IgM在感染初期出现，再次感染时产生量小。IgM抗体不能通过人的胎盘。IgM的这两种生物学特性使其对感染性疾病的诊断非常有用。患者血清中存在IgM表明近期受到感染，新生儿血清中存在IgM表明宫内感染。

 

 

【DESCREPTION】Immunoglobulin M (IgM) is a basic antibody that is produced by B cells. IgM is by far the physically largest antibody in the human circulatory system. It is the first antibody to appear in response to initial exposure to an antigen. The spleen, where plasmablasts responsible for antibody production reside, is the major site of specific IgM production. IgM is primarily found in serum, however, because of the J chain, it is also important as a secretory immunoglobulin.

 

IgM antibodies appear early in the course of an infection and usually reappear, to a lesser extent, after further exposure. IgM antibodies do not pass across the human placenta. These two biological properties of IgM make it useful in the diagnosis of infectious diseases. Demonstrating IgM antibodies in a patient\'s serum indicates recent infection, or in a neonate\'s serum indicates intrauterine infection.

 

 

【检测原理】本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠IgM单克隆抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品和检测样本进行孵育。洗涤去除未结合的物质后，加入辣根过氧化物酶标记的检测抗体孵育。洗涤后，加入显色底物TMB，避光显色。颜色反应的深浅与样本中IgM的浓度成正比。加入终止液终止反应，在450nm波长(参考波长570-630nm)测定吸光度值。

 

【PRINCIPLE OF THE ASSAY】This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for mouse IgM has been pre-coated onto a microplate. Standards and samples are pipetted into the wells for the first incubation. After washing away any unbound substances, a HRP-linked detect antibody specific for IgM is added to the wells. After washing, substrate solution is added to the wells and color develops in proportion to the amount of IgM bound in the initial step. The color development is stopped and the intensity of the color is measured.
