【背景介绍】粒细胞-巨噬细胞集落刺激因子(GM-CSF)是一种调节白细胞生长的细胞因子。GM-CSF刺激干细胞产生粒细胞(中性粒细胞、嗜酸性粒细胞和嗜碱性粒细胞)及单核细胞。此外，它对造血和非造血谱系细胞也具有活性。GM-CSF用作药物来刺激白细胞的产生，从而预防化疗后引起的中性粒细胞减少症。近来，人们正在临床试验评估GM-CSF作为一种潜在的疫苗佐剂对HIV感染患者的疗效。相关研究报道一些癌症(包括肺癌和急性髓细胞白血病)患者的血清中具有较高水平的GM-CSF，同样在支气管哮喘患者外周血和鳞屑提取物中也具有较高水平的GM-CSF。

 

【DESCREPTION】Granulocyte-macrophage colony stimulating factor (GM-CSF) is a cytokine that functions as a white blood cell growth factor. GM-CSF stimulates stem cells to produce granulocytes (neutrophils, eosinophils and basophils) and monocytes. In addition, it has additional activities on hematopoietic, as well as non-hematopoietic lineage cells.GM-CSF is used as a medication to stimulate the production of white blood cells and thus prevent neutropenia following chemotherapy. GM-CSF has also recently been evaluated in clinical trials for its potential as a vaccine adjuvant in HIV-infected patients. Increased levels of GM-CSF have been reported in the serum of some cancer patients, including patients with lung cancer and patients with acute myelogenous leukemia. GM-CSF has also been reported to be increased in the peripheral blood of bronchial asthma patients and in extracts of psoriatic scales.

 

【检测原理】本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人GM-CSF单克隆抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体，经过孵育，样本中存在的GM-CSF与固相抗体和检测抗体结合。洗涤去除未结合的物质后，加入辣根过氧化物酶标记的链霉亲和素(streptavidin-HRP)。洗涤后，加入显色底物TMB，避光显色。颜色反应的深浅与样本中GM-CSF的浓度成正比。加入终止液终止反应，在450nm波长(参考波长570-630nm)测定吸光度值。

 

【PRINCIPLE OF THE ASSAY】This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for human GM-CSF has been pre-coated onto a microplate. Standard, samples and biotin-linked detect antibody specific for GM-CSF are pipetted into the wells and GM-CSF present is bound by the immobilized antibody and detect antibody following incubation. After washing away any unbound substances, streptavidin-HRP is added. After washing, substrate solution is added to the wells and color develops in proportion to the amount of GM-CSF bound in the initial step. The color development is stopped and the intensity of the color is measured.
