产品描述Protein A is a 42 kDa surface protein originally found in the cell wall of the bacteria Staphylococcus aureus. It is encoded by the spa gene and its regulation is controlled by DNA topology, cellular osmolarity, and a two-component system called ArlS-ArlR. It has found use in biochemical research because of its ability to bind immunoglobulins. It is composed of five homologous Ig-binding domains that fold into a three-helix bundle. Each domain is able to bind proteins from many mammalian species, most notably IgGs (Table 1). It binds the heavy chain within the Fc region of most immunoglobulins and also within the Fab region in the case of the human VH3 family. Through these interactions in serum, where IgG molecules are bound in the wrong orientation (in relation to normal antibody function), the bacteria disrupts opsonization and phagocytosis. Recombinant Protein A is produced in E.coli and essentially functions the same as native Protein A.
Protein A may be conjugated with various reporter molecules, including fluorescent dyes (FITC), enzyme markers (peroxidase, b-galactosidase, and alkaline phosphatase), biotin, and colloidal gold without affecting the antibody binding site on the molecule. These conjugates are used to detect immunoglobulins in various immunochemical assays including Western blotting, immunohistochemistry, and ELISA applications.br> Protein A is conjugated to 10 nm colloidal gold. This gold conjugated protein A generates electron dense particles that are visible using electron microscopy.

羊抗牛IgG二抗广义上是指专门和一抗进行特异性反应和结合的抗体，在免疫学实验中，经常需要针对实验需求选择不同的二抗。通常情况下，某一特定的实验中可能同时有几种二抗可供选择，如何能选择到最适合该实验的二抗，需要综合考虑以下几个方面：
1、一抗的物种来源：一抗是什么物种来源的，相应的二抗也要是抗该物种的抗体。例如，如果一抗是在小鼠体内制备的，那么就应当选择抗小鼠的二抗，如果一抗是在兔体内制备的，那么二抗就应当是抗兔的抗体。至于二抗本身是在什么动物中制备的对二抗的质量并无明显的影响。
2、一抗的类别亚型：二抗需与一抗的类别或亚型相匹配。这通常是针对单克隆抗体而言。多克隆抗体主要是IgG类免疫球蛋白，因此相应的二抗就是抗IgG抗体。单克隆抗体的类别及亚型通常会在产品列表中列出，如果你的一抗是小鼠IgM，那么相应的二抗就应当是抗小鼠IgM，或是抗小鼠IgG抗体。如果单克隆一抗是小鼠IgG的某一亚型（IgG1，IgG2a，IgG2b，IgG3），那么几乎所有的抗小鼠IgG都可以与之结合，或者你也可以选择专门针对这一亚型的二抗，例如，如果你的一抗是小鼠IgG1，那么你可以选择抗IgG1的二抗，此种抗体在双标记实验中尤其适合。如果不知道一抗是哪一类别或亚型，那么抗小鼠IgG是一个不错的选择，因为此种抗体可以识别大多数类型的IgG免疫球蛋白。
3、二抗的偶联标记：一般来讲，偶联到二抗上的探针主要有HRP、荧光素、生物素等。选用哪种探针的二抗主要取决于具体的实验。ELISA、Western Blot和免疫组化中，最常用的二抗是酶标二抗（如HRP）；而免疫荧光和流式细胞术中通常使用荧光素标记的二抗，如FITC、CY3、PE等荧光素。
4、抗体的纯化方法：亲和层析纯化的抗体一般更受人们欢迎，因为这些产品的特异性较高，产生的非特异性条带和背景较低。但有些情况下也要考虑到纯化过程中IgG的损失，因为含有亲和力很高的抗体，尤其是当目的抗原存在的量很小的时候，高亲和性的IgG部分就显得尤为重要。
