传统DNA文库定量的标准流程耗费人力、时间，成本高，而单独采用常规定量的方法测量的是总的核算浓度，且灵敏度低、可信度差。最佳的簇密度或模板与磁珠的比率取决于合适的能进行PCR扩增的DNA分子的浓度，KAPA文库定量试剂盒正式通过高性能qPCR的方法对文库进行定量，减少簇密度或模板与磁珠比例的差异，同时减少传统标准流程的时间消耗、消除昂贵的滴定操作。

2、KAPA文库定量试剂盒流程

　　试剂盒包含：KAPA SYBR FAST qPCR、6个线性DNA标准品（10倍浓度稀释）、10×Primer Premix

1.将1ml的Primer Premix加入到5ml的KAPA SYBR FAST qPCR Master Mix(2x)中；

2.稀释dsDNA文库

用10mM Tris-HCI对文库进行1:1000倍稀释，如果需要可进行：1:2000,1:4000,1:8000稀释；

3.准备qPCR反应

20μl反应体系：12μl的KAPA SYBR FAST qPCR Master Mix（含Primer Premix）、4μl PCR水、4μl稀释的文库DNA或DNA标准品；

4.qPCR反应

起始变性5分钟，然后35个循环（95℃30秒、退火/延伸60℃45秒）；

5.数据分析

确认扩增效率位于90-110%之间；

6.计算文库浓度

根据说明书提供的公式推算绝对文库浓度；

7.根据计算所得的文库浓度推算未稀释的文库浓度，然后进行下一步实验。

传统qPCR试剂适用于短片段扩增，而对长片段、GC含量不均衡、复杂结构的片段扩增效率低，某些文库分子的定量不可靠。为了定量复杂DNA文库，KAPA特别改造了一种基于SYBR Green qPCR的酶，能有效扩增对野生型酶来说扩增困难的模板。KAPA文库定量试剂盒包含了该基因工程改造酶以确保能有效扩增长片段及GC含量跨度大的文库，对文库定量准确。

上图：稳定的扩增使不同文库qPCR定量准确KAPA文库定量试剂盒用于定量两个Illumina GA文库的浓度，这两个文库GC含量异常（Rhodococcus sp.～70%GC,如图，Staphylococcus sp.～35%GC未展示）。两个文库扩增效率均大于95%。样本连续两倍稀释（1:1000到1:16000），每个梯度3个重复，PCR反应参照产品使用说明书推荐参数。

可靠的DNA定量标准品，批次之间差异最小
