介绍：TriticumvulgarislectinorWeatgermagglutinin(WGA)isisolatedfromTriticumvulgaris(Weatgerm)andpurifiedbyaffinitychromatography.Thelectinhastwosubunitsandamolecularweightof36kDa.WGAselectivelybindstoN-Acetylglucosamine(GlcNAc)andtoN-acetylneuraminicacid(sialicacid)residuesofglycoproteinsandglycolipids.TheWGAlectinagglutinateserythrocytesandmosttypesofmalignantcells.Itagglutinatesrabbiterythrocytesat<0.1μg/mlfollowingtrypsintreatmentofthecells.Adding300mMN-Acetyle-D-Glucosaminegivesaninhibitionwithatiterthatisatleast8-foldlowerthanthecontrol.WheatgermagglutinininhibitstheC5areceptorinteraction,whichhasimplicationsinstudiesofreceptormicro-heterogeneityandligandbindingsites.WGAtogetherwithConAisthelectinmostwidelyusedasananalyticalandpreparativeagentwhenstudyingglycoproteinsandcellsurfaceproteins.Theimmobilizedlectincanbeusedforaffinitychromatographyofcellsandsub-cellularparticles.WGAissuppliedasawhitetopaleyellowlyophilizedpowderfromabuffercontaining10mMCH3COONH4.Nopreservativesareadded.ThepurityofWGAisdeterminedbySDS-electrophoresiswhichgeneratesthreebands,correspondingtothethreeisolectinsofWGA.Proteincontentisverifiedinspectrophotmetry.
