| 背景资料 | Suppressor of cytokine signaling 2 is a protein that in humans is encoded by the SOCS2 gene. This gene encodes a member of the STAT-induced STAT inhibitor (SSI), also known as suppressor of cytokine signalling (SOCS), family. SSI family members are cytokine-inducible negative regulators of cytokine signaling. The expression of this gene can be induced by a subset of cytokines, including erythropoietin, GM-CSF, IL10 and interferon-gamma (IFN-gamma). The protein encoded by this gene is found to interact with the cytoplasmic domain of insulin-like growth factor 1 receptor (IGF1R), and thus is thought to be involved in the regulation of IGF1R mediated cell signaling. By cytogenetic and radiation hybrid mapping, Yandava et al. (1999) mapped the SOCS2 gene to chromosome 12q21.3-q23. |

[list_product_images]Figure 1. Western blot analysis of SOCS2 using anti-SOCS2 antibody (PB0451). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: PC-12 Whole Cell LysateLane 2: SW620 Whole Cell LysateLane 3: HELA Whole Cell LysateLane 4: MCF-7 Whole Cell Lysate After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-SOCS2 antigen affinity purified polyclonal antibody (Catalog # PB0451) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for SOCS2 at approximately 22KD. The expected band size for SOCS2 is at 22KD.[/list_product_images]
