Blasticidin S (10mg/ml in Solution) 杀稻瘟菌素S （灭瘟素）作用机理

Blasticidin S是一种来自Streptomyces griseochromogenes的核苷类抗生素，通过干扰核糖体中肽键的形成来特异性地抑制原核和真核生物的蛋白质合成。Blasticidin S用于筛选携带有bsr或BSD耐受基因的转染细胞。杀稻瘟菌素具有快速而强效的作用模式，很低的抗生素浓度便能导致细胞迅速死亡。

Cat#60218ES10、60218ES50、60218ES60均已液体形式提供，浓度为10 mg/ml（in HEPES缓冲液，pH 7.5）。

Cat#:60218ES80为粉末形式提供，使用时可用无菌水或者缓冲液配制成5-10mg/ml的储存液。

目前已经克隆和测序的杀稻瘟菌素耐受基因有3种，一种是分离自产生菌Streptoverticillum sp.的乙酰基转移酶基因bls，另外两种是脱氨酶基因，包括从Bacillus cereus 中分离的bsr和从Aspergillus terreus 中分离的BSD 基因。Bsr和 BSD 基因是最常用的筛选标志，用于哺乳动物和植物细胞的基因转移实验，也可用于E.Coli。

应用浓度

1）Escherichia coli

E. coli对Blasticidin S的敏感性稍差，但是转化子对Blasticidin S具有耐受性，可以用低盐LB培养基（pH 8）进行筛选，浓度范围为50-100 μg/ml Blasticidin S。高pH值可以提高Blasticidin S的活性。

2）哺乳动物细胞
哺乳动物细胞中Blasticidin S的工作浓度范围在1-50 μg/ml。初次实验建议通过灭杀曲线来确定最佳使用浓度。

Blasticidin S (10mg/ml in Solution) 杀稻瘟菌素S （灭瘟素）操作步骤（哺乳动物稳转株筛选）

Blasticidin S通常使用浓度为10 μg/ml。携带bsr或BSD基因的质粒转染到细胞中，在含有Blasticidin S的正常生长培养基中孵育，用于筛选稳定转染细胞株。

（1）转染后48h，用含有适宜浓度Blasticidin S的新鲜培养基将细胞传代（注：细胞处于活跃分裂期时抗生素工作最好。细胞密度太高，抗生素效率降低。细胞分盘时覆盖率最好不超过25%）；

（2）每3-4天去除培养基，加入含抗生素的新鲜培养基；

（3）7天后检测细胞集落形成。根据宿主细胞种类和转染/筛选效率，集落形成可能需要增加一周或更久。（4）转移5-10个耐受克隆到35mm细胞盘中，加入选择培养基维持培养7天。随后用细胞毒性实验进行检测。
