【产品简介】

HelixGen MHC四聚体可应用于MHC Tetramer Assay(亦称Tetramer Assay或Tetramer Stain),能快速、简便地实现对抗原特异性T细胞(Antigen-specific T cells)的定性与定量分析。
大部分T细胞在其细胞表面表达单一的、高度特异性的抗原受体(TCR),能与MHC-抗原多肽复合物(MHC-antigen peptide complexes)结合并识别其中特异的抗原多肽,进而启动获得性/特异性免疫应答机制,比如T细胞介导的细胞免疫和B细胞介导的体液免疫。因此,抗原特异性T细胞在细胞免疫应答中发挥着核心作用,对其进行定性与定量分析有助于判定机体细胞免疫功能。由于TCR与MHC单体(MHC monomers)之间的亲和力低,相互结合后极易快速解离,重组的可溶性MHC-抗原多肽复合物单体并不适合用于抗原特异性的T细胞检测。1996年,斯坦福大学医学院John D. Altman博士等人开发出MHC四聚体技术:借助生物素-链霉亲和素级联反应放大系统,将MHC-抗原多肽复合物四聚化。MHC四聚体是由4个MHC-抗原多肽单体分子与1个标记有荧光染料的链霉亲和素分子组成。单个MHC四聚体可与同一T细胞表面的多个TCR结合,大大增强了MHC-抗原多肽复合物与TCR之间的亲和力和稳定性,提高了检测特异性,可通过流式细胞术成功实现对抗原特异性的T细胞的离体分析。

【产品用途】

适用于全血、外周血单核细胞(PBMCs)、细胞系等样本类型,能精确测定并可结合免疫磁珠分选技术(Immuno-magnetic sorting)高效分选抗原特异性的T细胞。
可应用于病原体免疫和疫苗研发、抗癌疗效评估、过敏监测与脱敏研究、自身免疫研究等领域。
