Introduction
组织因子途径抑制物（TFPI）是一种内源性蛋白酶抑制剂，它通过对组织因子/因子VIIa复合物的抑制而反馈抑制X a的生成，从而调节外源性凝血途径的启动（1）。TFPI含有一个阴性氨基端、三个串联的Kunitz蛋白酶抑制区，一个阳性羧基端。第一个Kunitz区是TF/VIIa复合物的结合部位，第二个Kunitz区是因子Xa的结合部位，由此生成的四重复合物TFPI/FXa/TF/FVIIa缺乏TF/VIIa 接触活性（2），第三个Kunitz区和羧基端介导与肝素和细胞表面（如内皮细胞）的结合（3）。TFPI主要由内皮细胞合成，在人体有三个池：血小板上的TFPI约占10%、血管内皮细胞腔面结合的TFPI、血浆中游离的或与脂蛋白结合的TFPI（4）。血浆中的TFPI主要有34和40 kDa两种形式，浓度约为50~100ng/ml（5，6）。在血栓形成、动脉硬化和肝素化等的研究中检测TFPI有重要意义，血浆高水平的TFPI可见于老年人、怀孕妇女及进展期肿瘤患者（7，8，9）。

Principle of the Assay
The AssayMax Human Tissue Factor Pathway Inhibitor (TFPI) ELISA kit is designed for detection of human TFPI in plasma, serum, milk, and cell culture samples. This assay employs a quantitative sandwich enzyme immunoassay technique that measures TFPI in less than 4 hours. A polyclonal antibody specific for TFPI has been pre-coated onto a 96-well microplate with removable strips. TFPI in standards and samples is sandwiched by the immobilized polyclonal antibody and biotinylated polyclonal antibody specific for TFPI, which is recognized by a streptavidin-peroxidase conjugate. All unbound material is then washed away and a peroxidase enzyme substrate is added. The color development is stopped and the intensity of the color is measured.
