检测原理：

欣博盛 QuantiCyto® ELISA试剂盒采用双抗体夹心法：抗人IL-1β单抗包被于酶标板上，标本和标准品中的人IL-1β会与单抗结合，游离的成分被洗去。加入生物素化的抗人IL-1β抗体和辣根过氧化物酶标记的亲和素，生物素与亲和素特异性结合；抗人IL-1β抗体与结合在单抗上的人IL-1β结合而形成免疫复合物，游离的成分被洗去。加入显色底物，若反应孔中有人IL-1β，辣根过氧化物酶会使无色的显色剂现蓝色，加终止液变黄。在450 nm处测OD值，人IL-1β浓度与OD450值之间呈正比，可通过绘制标准曲线求出标本中人IL-1β的浓度。

人IL-1β简介：

人IL-1β为IL-1家族的成员之一，IL-1家族包括IL-1α、IL-1β、IL-1ra、IL-18及IL-1F5至F10。所有家族成员都具有12β链，β三叶草构像。所有家族成员被认为是从同一祖始基因经多次复制而来。IL-1α和IL-1β为不同基因的产物，虽其氨基酸 序列只有大约25%的同一性，但它们识别相同的细胞表面受体。多种细胞在应答刺激如炎性因子、感染或微生物内毒素等时，产生这两种蛋白质。这些细胞因子可以增强内皮细胞表达粘附分子，从而使白血细胞迁移至感染位点，重建下丘脑体温调节中心导致体温升高，表现为发热症状。因此IL-1 亦被称为体内致热原。体温升高有助于机体免疫系统抵抗病原物。IL-1在调控血细胞生成方面也发挥着重要作用。
