协议摘要
 

1. 制备密度为 0.5 - 1 × 10 的细胞6细胞/毫升
2. 用测试化合物处理细胞以诱导超氧化物
3. 将 1 μL 500X MitoROS™ 580 加入 0.5 mL 细胞悬液中
4. 将细胞在 37°C 下染色 1 小时
5. 使用带FL2通道的流式细胞仪监测荧光强度

重要说明
在开始实验之前，请在室温下解冻所有组分。

储备液的制备
除非另有说明，否则所有未使用的储备溶液应分成一次性等分试样，并在制备后储存在-20°C。避免反复冻融循环。

1. MitoROS 580储备溶液（500X）：
将100μLDMSO（组分C）加入MitoROS 580（组分A）的小瓶中，混合均匀，制成500X MitoROS™™™ 580储备溶液。避光。注意：储存时，请密封管子。


实验方案样本

1. 对于每个样品，在您选择的 0.5 mL 生长培养基或缓冲液中制备细胞，密度为 5×10⁵至 1×10⁶细胞/毫升。注意：应单独评估每个细胞系，以确定超氧化物诱导的最佳细胞密度。

 

1. 在检测缓冲液（组分 B）或所需缓冲液（如 PBS）中用 25 μL 20X 测试化合物处理细胞以诱导超氧化物。对于对照细胞（未处理的细胞），加入相应量的复合缓冲液。

 

1. 将细胞在37°C下孵育至少30分钟或所需的时间，避光。注意：用50μM抗霉*A（AMA）在37°C下处理Jurkat细胞30分钟以诱导超氧化物。有关详细信息，请参见图 1。化脓花青素（50μM）或H₂O₂（1 mM） 也可用于诱导超氧化物。

 

1. 将 1 μL 500X MitoROS™ 580 储备溶液加入 0.5 mL 细胞悬液中。

 

1. 在 37°C 下孵育 1 小时。注意：对于贴壁细胞，用0.5 mM EDTA轻轻提起细胞以保持细胞完整，并在用MitoROS™ 580孵育之前用含血清的培养基洗涤细胞一次。适当的孵育时间取决于所使用的单个细胞类型和测试化合物。优化每个实验的孵育时间。

 

1. 使用具有FL2通道的流式细胞仪（Ex / Em = 488 / 590 nm）监测荧光强度。对感兴趣的细胞进行门控，不包括碎片。
