Rh110 Factor Xa 检测试剂盒提供了一种方便的检测方法，用于筛选 FXa 抑制剂或使用荧光底物连续检测酶活性。在 FXa 蛋白酶切割后，该底物会产生 Rh110（罗丹明 110）荧光团，该荧光团具有亮绿色荧光，可在激发/发射 = 490 nm/520 nm 时检测到。Rh110 的更长波长光谱和更高消光系数提供更高的灵敏度和更少的其他反应组分的干扰。检测限：0.31 ng/ml
Xa 因子 (FXa) 是由两个二硫键连接的亚基组成的丝氨酸内肽酶。FXa 通过凝血酶原酶复合物将凝血酶原转化为凝血酶，从而导致血栓形成。FXa 由酶原因子 X 通过内在和外在途径产生，是凝血酶生成传播的限速步骤。在 Ca2+ 离子存在的情况下，FXa 与活化血小板的磷脂膜上的凝血酶原酶 Va 形成。
Add test compounds and diluted enzyme solution to the microplate wells. The suggested volume of enzyme solution for one well of a 96-well plate is 40 uL and test compound is 10 uL.
Simultaneously set up the following control wells, as deemed necessary:
Positive control contains the diluted Factor Xa without test compound.
Inhibitor control contains the diluted Factor Xa and inhibitor.
Vehicle control contains enzyme and vehicle used in delivering test compound (e.g. DMSO, concentration not to exceed 1%).
Test compound control contains 1X assay buffer and test compound. Some test compounds have strong autofluorescence and may give false results.
Substrate control contains 1X assay buffer.
Using the assay buffer, bring the total volume of all controls to 50 uL.
Optional: Pre-incubate the plate for 10 min. at assay temperature. Any temperature (the assay temperature) from room temperature to 37 ℃ may be used, as long as the subsequent incubations are performed at the same temperature.
